Since the discovery of restriction endonuclease, genetic engineering has been able to manipulate DNA. Many hundred restriction enzymes are now known, each having a unique sequence requirement that determines where it can cut DNA. These were isolated from bacterial cells and are useful for molecular biologists. Therefore, using a restriction enzyme to digest DNA results in a specific collection of fragments that can be separated by electrophoresis and then examined.
